dc.description.abstractIn this study, the in vitro sterile plantlets and callus production in Catharanthus roseus L., which is rich in the number of alkaloid and arouses interest especially in cancer treatments, the multiple shoot production from sterile plantlets and the alkaloid analysis were made in both productions. C.roseus seeds were firstly germinated in full-power MS medium, the hypocotyl, cotyledon and root explants were promoted to callus production in different plant growth regulators supplemented basic MS medium. The dominant presence of a class of alkaloid (tryptophan) was identified in the 0. 1 mg L-1 kinetin+ 1 mg L-1 NAA and 0.1 mg L-1 kinetin+ 2 mg L-1 2.4 D supplemented nutrition environment. In the study, the plantlets (root and hypocotyl + cotyledon) germinated in 1/2 MS medium were also separated from their roots and were cultivated in basic MS medium supplemented with different concentrations of BA for promotion of multiple shoot growth. To control alkaloid analyses were made for in vivo grown C.roseus plant (provided during the flowering period). The best shoot growth was seen in 1 mg L-1 BA supplemented nutrition environment and the obtained serpentine indol alkaloid was determined to be higher than the control plant although alkaloids obtained from the shoots grown in that nutrition environment were the same compounds as the control plant. Additionally, tryptamine alkaloid occurred only in control and the shoots obtained from that nutrition environment. © 2017, Centenary University. All rights reserved.en_US
